While he was in New York, Horkheimer's essays "The Latest Attack on Metaphysics" and "Traditional and Critical Theory," which would soon become instructive for the institute's self-understanding, were the subject of intense discussion. Soon after his return to Europe, Gretel moved to Britain, where she and Adorno were married on 8 September 1937. A little over a month later, Horkheimer telegrammed from New York with news of a position Adorno could take with the Princeton Radio Project, then under the directorship of the Austrian sociologist Paul Lazarsfeld. Yet Adorno's work continued with studies of Beethoven and Richard Wagner (published in 1939 as "Fragments on Wagner"), drafts of which he read to Benjamin during their final meeting, in December on the Italian Riviera. According to Benjamin, these drafts were astonishing for "the precision of their materialist deciphering" as well as the way in which "musical facts ... had been made socially transparent in a way that was completely new to me." == Engineering of 4S pathway enzymes == The desulfurization rate for the wild-type 4S pathway enzymes is low when compared to the rate that needs to be achieved for a viable application in the industrial sector. An increase of 500-fold on the overall rate of the pathway is the required improvement for an efficient application of this biodesulfurization method. Directed evolution, rational design or a combination of both strategies are some of the approaches that have been applied to tackle the lack of catalytic efficiency and stability of the 4S enzymes. The 4S pathway best improvement to date was obtained by a directed evolution approach in which Rhodococcus strains were transformed with a plasmid encoding a modified dsz operon (which encodes for DszA, DszB and DszC). After 40 subculturing events in a medium in which DBT was the sole sulfur source, the modified Rhodococcus strains presented a 35-fold improvement. The strong feedback inhibition of DszC was also tackled by a combination of directed evolution and rational design approach to desensitize DszC to the 4S pathway product, HBP. The bacterial strain expressing the DszC A101K mutant showed higher activity relative to the wild-type strain. Additionally docking of HBP to the protein revealed that HBP forms a π-interaction with Trp327, thus inhibiting DszC. The A101K/W327C (AKWC) double mutant revealed to be desensitized to low HBP concentrations and the bacterial strain expressing the AKWC DszC was 14-fold more efficient than the wild-type strain.

In biology, a pair bond is the strong affinity that develops in some species between a mating pair, often leading to the production and rearing of young and potentially a lifelong bond. == Antibiotics == PBPs bind to β-lactam antibiotics because they are similar in chemical structure to the modular pieces that form the peptidoglycan. When they bind to penicillin, the β-lactam amide bond is ruptured to form a covalent bond with the catalytic serine residue at the PBPs' active site. This is an irreversible reaction and inactivates the enzyme. There has been a great deal of research into PBPs because of their role in antibiotics and resistance. Bacterial cell wall synthesis and the role of PBPs in its synthesis is a very good target for drugs of selective toxicity because the metabolic pathways and enzymes are unique to bacteria. Resistance to antibiotics has come about through overproduction of PBPs and formation of PBPs that have low affinity for penicillins (among other mechanisms such as lactamase production). These experiments change the structure of PBP by adding different amino acids into the protein, allowing for new discovery of how the drug interacts with the protein. Research on PBPs has led to the discovery of new semi-synthetic β-lactams, wherein altering the side-chains on the original penicillin molecule has increased the affinity of PBPs for penicillin, and, thus, increased effectiveness in bacteria with developing resistance. Presence of the protein penicillin binding protein 2A (PBP2A) is responsible for the antibiotic resistance seen in methicillin-resistant Staphylococcus aureus (MRSA).
