=== Absorbance === When components are detected optically, the mobile phase should have high transmittance at the wavelength the detector uses. Most solvents are more transparent at longer wavelengths. Consequently, researchers usually speak of "cutoff wavelengths" of solvents. A solvent's cutoff wavelength is the wavelength at which the absorbance of the solvent equals 1 AU. A modifier should only be used with light above the cutoff wavelength. == Causes == Colombia has a long history of political violence. Land, power, and wealth were unevenly distributed and many rural citizens were used to having to fend for themselves. There is no consensus about the date on which the conflict began, with some saying 1958 (with the start of the Frente Nacional (National Front)) and others 1964 (with the creation of the FARC). In the mid-1980s, Colombia granted local governments greater political and fiscal autonomy. This strengthened the government's position in more remote regions. In 1985, during peace talks with then-President Belisario Betancur, the FARC created the left-wing Patriotic Union party as a route from violence to mainstream politics. Between 1985 and 2002, 4,153 members and supporters of the party were kidnapped and murdered by right-wing paramilitaries with government support. This included two presidential candidates, 6 out of 16 congressmembers, 17 regional representatives, and 163 councilmembers. These killings aggravated the conflict. In the 1980s, drug trafficking increased, bringing a concomitant increase in violence. Trafficking began in the 1960s and 70s when a group of Americans began to smuggle marijuana. Later, the American Mafia moved into drug trafficking in Colombia alongside local marijuana producers. Cocaine and other drugs produced in Colombia were mostly consumed in the US, as well as Europe. Organized crime in Colombia grew increasingly powerful in the 1970s and 80s with the introduction of massive drug trafficking to the United States from Colombia.
== Drug pricing issue and exit in the Swiss market == In July 2025, Roche withdrew Lunsumio from the Swiss market following failed price negotiations with the Swiss Federal Office of Public Health (BAG). The drug had been available since February 2023 through a special "Early Access" program that allowed reimbursement before complete clinical data was available. Under this arrangement, Roche initially covered treatment costs until efficacy was demonstrated, after which the remaining therapy could exceed 100,000 Swiss francs. When Swissmedic extended the temporary authorization in February 2025 due to still-missing complete clinical data from Phase III studies, Roche rejected the previously negotiated pricing model and demanded higher prices. The BAG refused to modify the agreement, citing concerns that exceptions would set a precedent for other pharmaceutical companies and demanding additional efficacy evidence despite the drug showing positive effects in Phase II studies. According to Swissmedic pricing data, a single 60-mg dose (two vials) of Lunsumio costs over 7,000 Swiss francs. === Protocol example === A ZooMS protocol (Fig. 1) typically consists of an extraction, denaturation, digestion and filtration step, followed by mass spectrometric analysis. Various destructive and non-destructive extraction protocols have already been discussed in some detail above. The key is to extract the protein preserved in the sample and then bring it into solution, usually an ammonium bicarbonate buffer. Denaturation is done to unfold the proteins and make them more accessible for the enzymatic digestion. It is done by heating the solubilised sample at around 65 °C. Then an enzyme, trypsin, is added to the solution. Trypsin cleaves the protein after every arginine or lysine amino acid in its sequence, resulting in peptide fragments of predictable masses. After digestion the sample is filtered with C18 filters to get rid of non-proteinaceous material and the sample is now ready for mass spectrometric analysis, which for ZooMS generally means MALDI-TOF MS.
